New Genetic Alterations Associated With Human Height Identified

A large collaborative study has added to the growing list of genetic variants that determine how tall a person will be. The research, published on December 30 in the American Journal of Human Genetics, identifies uncommon and previously unknown variants associated with height and might provide insight into the genetic architecture of other complex traits.

Although environmental variables can impact attained adult height, it is clear that height is primarily determined by specific alleles that an individual inherits. Height is thought to be influenced by variants in a large number of genes, and each variant is thought to have only a small impact on height. However, the genetics of height are still not completely understood. "All of the variants needed to explain height have not yet been identified, and it is likely that the additional genetic variants are uncommon in the population or of very small effect, requiring extremely large samples to be confidently identified," explains Dr. Hakon Hakonarson from The Children's Hospital of Philadelphia.

To search for genetic variants associated with adult height, researchers performed a complex genetic analysis of more than 100,000 individuals. "We set out to replicate previous genetic associations with height and to find relevant genomic locations not previously thought to underpin this complex trait" explains Dr. Brendan Keating, also from The Children's Hospital of Philadelphia. The authors report that they identified 64 height-associated variants, two of which would not have been observed without such a large sample size and the inclusion of direct genotyping of uncommon single-nucleotide polymorphisms (SNPs). A SNP is a variation in just one nucleotide of a genetic sequence; think of it as a spelling change affecting just one letter in an uncommonly long word.

These results suggest that genotyping arrays with SNPs that are relatively rare and occur in less than 5% of the population have the ability to capture new signals and disease variants that the common SNP arrays missed (i.e., 30 new signals in this study), as long as sample sizes are large enough. These low-frequency variants also confer greater effect sizes and, when associated with a disease, could be a lot closer to causative than more common variants. "The increased power to identify variants of small effect afforded by large sample size and dense genetic coverage including low-frequency SNPs within loci of interest has resulted in the identification of association between previously unreported genetic variants and height," concludes Dr. Keating.

Eating Poorly Can Make You Blue: Trans-Fats Increase Risk of Depression, While Olive Oil Helps Avoid Risk

Researchers from the universities of Navarra and Las Palmas de Gran Canaria have demonstrated that the ingestion of trans-fats and saturated fats increase the risk of suffering depression, and that olive oil, on the other hand, protects against this mental illness.

They have confirmed this after studying 12,059 SUN Project volunteers over the course of six years; the volunteers had their diet, lifestyle and ailments analyzed at the beginning of the project, over its course and at the end of the project. In this way the researchers confirmed that despite the fact that at the beginning of the study none of the volunteers suffered from depression, at the end of the study 657 new cases had been detected.

Of all these cases, the participants with an elevated consumption of trans-fats (fats present in artificial form in industrially-produced pastries and fast food, and naturally present in certain whole milk products) "presented up to a 48% increase in the risk of depression when they were compared to participants who did not consume these fats," affirmed Almudena Sánchez-Villegas, Associate Professor of Preventive Medicine at the University of Las Palmas de Gran Canaria, first author of the article.

In addition, the study demonstrated a dose-response relationship, "whereby the more trans-fats were consumed, the greater the harmful effect they produced in the volunteers," the expert stated.

Furthermore, the team, directed by Miguel Ángel Martínez-González, Professor of Preventive Medicine at the University of Navarra, also analyzed the influence of polyunsaturated fats (abundant in fish and vegetable oils) and of olive oil on the occurrence of depression. "In fact, we discovered that this type of healthier fats, together with olive oil, are associated with a lower risk of suffering depression," emphasized the researcher and director of the SUN Project.

150 million persons depressed worldwide

Thus, the results of the study corroborate the hypothesis of a greater incidence of the disease in countries of the north of Europe compared to the countries of the south, where a Mediterranean dietary pattern prevails. Nevertheless, experts have noted that the incidence of the disease has increased in recent years, so that today some 150 million persons are affected worldwide, where it is the principal cause of loss of years of life in those countries with a medium-to-high per capita income.

This due, according to Almudena Sánchez Villegas, "to radical changes in the sources of fats consumed in Western diets, where we have substituted certain types of beneficial fats -- polyunsaturated and monounsaturated in nuts, vegetable oils and fish -- for the saturated and trans-fats found in meats, butter and other products such as mass-produced pastries and fast food."

In addition, the research -- published in the online peer reviewed journal PLoS ONE -- has been performed on a population with a low average intake of trans-fats, given that it made up only 0.4% of the total energy ingested by the volunteers. "Despite this, we observed an increase in the risk of suffering depression of nearly 50%. On this basis," concluded Miguel A. Martínez, "we derive the importance of taking this effect into account in countries like the U.S., where the percentage of energy derived from these foots is around 2.5%."

Finally, the analysis, headed by the University of Navarra and the University of Las Palmas de Gran Canaria, suggests that both depression as well as cardiovascular disease are influenced in a similar manner by diet, and might share similar mechanisms in their origin. This hypothesis is further suggested by numerous studies that indicate the harmful effect of trans-fats and saturated fats on the risk of cardiovascular disease.

Scientists Unlock the 'Gates' on Sudden Cardiac Death

Australian researchers have come one step closer to understanding how the rhythm of the heartbeat is controlled and why many common drugs, including some antibiotics, antihistamines and anti-psychotics, can cause a potentially fatal abnormal heart rhythm.

It is estimated around 40-50% of all drugs in development will block one of the main 'channels' that carries electricity in the heart and, as a result, can cause heart rhythm problems called cardiac arrhythmias. Most sudden cardiac deaths are caused by cardiac arrhythmias.

Since 1996, nine drugs have been withdrawn from the market or had their use severely restricted due to this serious side effect.

In a paper published in this month's edition of the journal Nature Structural and Molecular Biology, Victor Chang Institute in Sydney have discovered a key clue as to why this happens, by understanding how the 'gates,' which effectively 'open' and 'close' the channel, operate.

"Just like a set of metal wires that carry electricity to light up our streets, our body has a series of channels that carry tiny charged particles called ions, into and out of cells, to trigger a heartbeat," said Professor Jamie Vandenberg, Head of the Cardiac Electrophysiology Laboratory at the Victor Chang Institute.

"Depending on the position of these gates, many common drugs bind, or attach themselves to these channels, blocking the ions from passing through. This causes what we call Long QT syndrome, where the length of the heart beat is longer than usual, which greatly increases the risk of arrhythmia."

The group of drugs most commonly associated with this side effect are anti-psychotic drugs, taken by patients with schizophrenia and other psychiatric disorders. Patients taking these drugs are up to three times more likely to die of sudden cardiac death due to an abnormal heart rhythm.

The team of researchers, led by Professor Vandenberg, studied the hERG potassium channel, an ion channel that determines how long each heart beat lasts and the channel which is most susceptible to being 'blocked' by drugs.

"The hERG channel is a particularly 'sticky' channel, in that most drugs will bind to it when the outer gate is closed. What we've done is to discover how these outer gates operate, in the hope that we can then design drugs more effectively to minimise the unwanted side effects," said Professor Vandenberg.

"The gates to this channel operate in a much more complex way than was previously thought -- much like a Japanese puzzle box, they require a series of complicated, interrelated movements to open them. It is not simply a matter of lifting and shutting a lid as was commonly believed," says Professor Vandenberg.

The team suspects this 'gate mechanism' will also apply to other channels that are important in the heart's electrical system, as well as those that control electrical communication in the brain.

"The biggest benefit of this research is that it should allow the better design of drugs so they no longer block these important electrical channels in the heart," added Professor Vandenberg. "In time, this should allow patients the freedom and peace of mind to take their medication without the fear of their heart suddenly stopping."

Success in “Best cars of 2011” survey: Audi takes top honors in four categories

Again this year, the Audi success story continues in the “Best cars of 2011” reader survey, conducted by Germany’s specialist car magazine “auto motor und sport” (ams). Readers awarded top honors to four Audi models – the Audi A1, the A4/A5, the R8 Spyder and the Audi Q5. The second-ranked Audi A8 and the third-place A3, A6 and R8 Coupé round out the outstanding result.
The four first-place awards make Audi the most successful brand in this year’s survey. Markets ponder the prospect of strong US growth.

FX markets remain fairly range bound as we wait for the US fourth quarter GDP due out at 1330 GMT. The data should set the tone for the dollar as we end the week. The US economy is expected to have expanded by 3.5 per cent in the final three months of the year, according to a survey of the market by Bloomberg. The balance of risks is to the upside and growth could come in above 4 per cent. Trade should have a positive impact on the US economy after subtracting from growth in both the second and third quarters of 2010. The trade balance stabilised just below $40bn in December and exports have been growing. Added to this, the extension of the Bush-era tax cuts has boosted consumer confidence and suggests that retail trade will pick up albeit from a low level.

The ingredients are in place for the US economy to pick up after disappointing data in the second and third quarters of 2010. If growth is at 3.5 per cent, then the US economy would be back to its pre-recession size. While that could boost investor optimism in the short-term, it has taken a long-time to bounce back from this recession compared to previous downturns and unemployment still remains high. The output gap is still sizable due to the economic potential that was lost as a result of the financial crisis' devastating economic impact. This is keeping inflation and unemployment at disappointing levels. Indeed, The Fed's preferred measure of consumer prices the core PCE will also be released for the fourth quarter along with the GDP data later today. This is expected to have fallen to 0.4% in the fourth quarter from 0.5% in the third, which is the lowest level for more than two decades.

So while an upside surprise in growth data may spur the dollar to a strong finish for the week as it starts trading as a growth currency, the actual outlook for the economy is slightly more subdued. Treasury Secretary Timothy Geithner is talking at the economic forum in Davos said that the US is not going to experience a boom and growth is not rapid enough to bring down unemployment. Without monetary and fiscal stimulus the US economy would still be languishing in the doldrums. This support also raises questions about the US deficit. Debt levels are expected to exceed more than $14 trillion this year larger than the entire US economy, for the first time. A credit rating agency warned that it may need to place a negative outlook on the US's Aaa credit rating sooner than anticipated as the budget deficit widens. This is a serious issue that the US needs to address. While the US's debt levels are in focus, the USD may find it hard to sustain a rally even if growth is strong.

There was more bad news for Spain's economy. The unemployment rate is rising again; it topped 20 per cent in the fourth quarter of 2011, up from 19.79 per cent in the third quarter. However, the country is taking steps to sort out its banking sector. Although profit fell at La Caixa, a domestic savings bank after an increase in bad loans, it announced plans to list on the stock exchange. It is the first of Spain's domestic lenders to list, which is part of the government's plans for re-capitalising the troubled domestic lenders, which make up 50 per cent of Spain's banking sector. This is the first step on a very long road to rehabilitation, but this along with some hawkish comments from ECB members is helping EURUSD to sustain gains above 1.3700 as we lead into the GDP data this afternoon.

Elsewhere, the Swiss franc is performing well after a leading economic indicator for January surpassed market expectations. The yen has also pared some of yesterday's losses after the S&P downgrade to Japan's sovereign credit rating.

Stocks are fairly directionless in the absence of much economic data out of Europe. A close above 1,300 in the S&P 500 this week would be a bullish sign for further gains. Strong US growth may fuel risky assets during the New York session. Money supply in the Eurozone was the highlight. It remains fairly lacklustre, and loan growth eased at the end of last year. The annualised rate of money growth was 1.7 per cent in December, which may reduce pressure to raise interest rates in the currency bloc as this is one of the key measures used by the ECB to try and gauge future inflation

For example, when I am contacted by a prospective client, I have them fill out a website requirements document that poses various questions to help me better understand what they are looking for in a website. Some of the questions I ask are:
Describe the nature of your business.
  • Who is your target audience?
  • What is the background on the project? (Is it brand new? Has it already begun?)
  • What are the goals and objectives of the project?
  • What is the timeline for the project?
2. Communicate Professionally
Your professionalism can win you contracts, and your communication skills add to the complete package. Take time to proofread all emails prior to sending; use a business email address with a proper signature; answer the phone professionally; and speak articulately and competently at all times.
While my email signature has evolved over time, below is the general format I follow, which has worked well for me:

Client meetings, even those that take place over the telephone, are an integral part of every successful business. Follow these tips to make your meetings as productive as possible.
3. Schedule and Prepare Thoroughly
We’re all busy these days, so scheduling your meetings in advance ensures that you and your clients have an adequate amount of uninterrupted time to speak. Once your meeting is scheduled, take time to prepare an agenda that outlines focus points and sets a structure. Sharing the agenda for the meeting gives both you and the client an opportunity to fully prepare.
Because you may not be using the same calendar or scheduling program as your client, confirming the date and time of your meetings in an email and sending a reminder and the agenda the day before is good practice. If you are unsure how to format an agenda, plenty of templates are available for free online.
4. Speak, Pause, Listen
When you have several topics to tackle, rushing through them to get all of your ideas out may be tempting. But this causes confusion and makes the client feel that their input is not important. Slow down, and remember that communication is a two-way street. Establish a give-and-take that allows both parties to have their say.
One way to become a better listener is to limit or eliminate distractions during your conversations. That may mean closing your email client, turning off the television and closing the door to your office. By doing these small things, you ensure that the client has your full attention, and they will sense that, too.
5. Follow Up in Writing
While you may be taking notes during phone or in-person meetings, the other party might not be, so follow up after the meeting with a written message, giving an overview of the discussion to make sure you are both on the same page. Summarize what was agreed, repeat questions that were raised and outline the next steps and responsibilities for both parties.
In addition to sending your notes, invite the other party to give their feedback on what you have sent. This way, it becomes a collaborative document and not just one person’s view.
Customer Service
Your clients want to feel that they are your priority. You can make them feel so by providing exemplary customer service. Try these communication-focused actions to improve your customer service.
6. Ask for Feedback
One way to maintain long-term relationships with your clients is by keeping open lines of communication. This means asking them for their input on how things are going and how they feel about the service you’re providing. This can be accomplished by inquiring at the end of a project, during day-to-day conversations or through formal surveys. The format matters less than the actual act of it, so work it into your business and fine tune as you go along.

7. Address Problems
If a client is unhappy, don’t ignore their complaints. Ask them why they are unhappy and what you can do to fix the situation. The longer you wait to bring it up, the worse it will get. Addressing the issue and being accountable when appropriate puts you on the path to resolution. And your willingness to face the problem head-on tells the client that you care about the project and their satisfaction.
If a client complains about your turnaround time or responsiveness, you may need to create a more formal project plan to clarify expectations. A working document like this can also eliminate some of the uncertainty regarding responsibilities and keep everyone on track.
8. Try a New Format
If a problem with your client stems from miscommunication, try a different method of communication. If you have been handling everything via email, schedule a phone call to see if that clears things up. After the call, you can summarize the conversation in an email to the client, which will give you another opportunity to get both of you on the same page again.
Today, so much communication is done via email that the opportunity for major miscommunication is almost inevitable. A rule of thumb is to limit your email to one screen-full (i.e. above the fold); anything that requires more space than that should be handled by phone. This should help you avoid some of the pitfalls of relying on email alone.
Face-to-Face Networking
Networking events, conferences and other face-to-face opportunities can take your business to a new level. These tips focus on helping you get the most from in-person networking activities.
9. Communicate Confidently
Be confident and use body language to support that confidence. Shake hands firmly, smile and make eye contact while communicating at live networking events. Don’t forget to bring business cards to hand out to everyone you meet, and remember to relax and be yourself.
Before heading out to a networking event, practice introducing yourself to new people to gain confidence. Working on your introduction with someone you trust and asking for their feedback also helps.
10. Prepare an Elevator Speech
An elevator speech helps you make the most of a first impression, while making networking situations easier and more productive. Be prepared with your speech and ready to answer common questions about your business and what you do. Practice your elevator speech ahead of time so that you are relaxed and comfortable with introducing yourself.
Your elevator speech should last no longer than 30 seconds and should convey how your product or service solves a problem for your target audience. An elevator speech could go something like:

Whether you market your business online, in person or through traditional advertising, communication is key to brand awareness. Here are two secrets to magnify the impact of your marketing across the board.
11. Be Responsive
A big part of marketing is being available to your target audience and following up when necessary. If you market your business through social media outlets—including Twitter, Facebook and blogging—watch for and respond to comments, questions and especially complaints. And when you are contacted as a result of offline marketing activities, respond quickly and professionally.
Plenty of recent examples on Twitter show how certain brands have been slow to respond to criticism, hoping it would die down, only to see it spin out of control. Also, when you do respond on social media websites, keep it professional, and avoid confrontation because that would only spread the fire.
12. Write Well
You can’t successfully promote your business if your marketing copy is not clear, concise and action-provoking. If writing is not your forte, consider hiring someone to help you craft copy that attracts potential clients, generates interest in your services and motivates potential clients to action.
To strengthen your writing skills, start a swipe file of marketing copy that you like and have found inspirational. Read through it and make notes of what you like in particular and what pulls you in. By making this a frequent exercise, you should be able to learn what makes good copy good and bad copy bad.

Pakistan, China signed 16 pacts since 2007


ISLAMABAD  (January 30, 2011) : As many as 16 agreements were signed between Pakistan and China since 2007. According to information provided to the National Assembly Secretariat, an agreement was signed on establishment of Joint Investment Company between Government of Pakistan and China Development Bank (CDB) on July 18, 2007.

Another Grant Assistance Agreement on Economic and Technical Co-operation was signed with China Exim Bank on April 07, 2007 In 2008, two agreements were signed with China including Preferential Buyer's Credit Loan Agreement for Improvement of KKH and Grant Assistance Agreement on Economic and Technical Co-operation signed on January 18, 2008 and on October 15, 2008 with China Exim Bank respectively.

Similarly, five agreements were signed with China in 2009, Preferential Buyer's Credit loan agreement on Roads & Bridges (Package-I) under AJK development programme signed on June 18, 2009. Preferential Buyer's Credit loan agreement Urban Infrastructure Development (Package-II) under AJK Development Programme signed on June 18, 2009. Government concessional loan agreement on Pak IR Satellite system project signed on October 15, 2009. Buyer Credit loan agreement on Procurement and manufacturing of 75-Deisel Engine Locomotives. Buyer Credit loan agreement on procurement and manufacturing of 202 Rails Coaches signed on December 14, 2009.

In 2010, Government Concessional Loan Agreement Chashma-3 & 4 units Project signed on February 22, 2010. Preferential Buyer Credit Loan Agreement of Chashma-3 & 4 units Project signed on February 22, 2010. Buyer Credit of Chashma-3 & 4 units Project signed on April 09, 2010. Grant Assistance Agreement on Economic and Technical Co-operation for Flood Relief signed between Government of Pakistan and China Exim bank on July 07, 2010.

Similarly, Concessional Loan Agreement of Pak-Sat-1R Ground Control Segment project signed on October 27, 2010. Concessional Loan with China Exim bank on procurement of Non intrusive vehicle X-Ray system project signed on December 17, 2010 and Concessional Loan Agreement on Safe City Islamabad project signed with China Exim Bank on December 17, 2010.
